The sequencing of the genome of many animal and plant species provides access to ever more complete databases.
To be able to achieve this level of research, it was necessary to clearly understand gene expression at a specific time and place.
Immunohistochimie – IHC

Immunohistochemistry (IHC) is a method of locating proteins (antigens) in the cells of a tissue section or sample by detecting antigens with antibodies.

This technique exploits specific Antigen/Antibody bonds in biological tissues. Antibodies can be of polyclonal or monoclonal origin, monoclonal antibodies being more specific in nature.

IHC can be applied in many different ways by using different probes and developers. Indeed, primary antibodies can be coupled with substrate reactive enzymes, or coupled to fluorophores and visualized under an epifluorescence microscope. This last variation of IHC is often referred to as immunofluorescence.

To finely analyze the location of gene activation zones, the use of Antibody and IHC probes is very important. However, these manual techniques are difficult to implement, particularly on in toto samples such as a tissue fragment or animal embryo.

Thanks to the Flo400, the samples are subjected to a permanent flow of solvents that guarantees the transfer of the solution to the nuclei of the cells. A constant supply of reagent avoids any stagnation and preserves the integrity of biological samples.
The constant recycling of solvents, RNA probes and antibodies considerably reduces the analysis time. The solutions collected at the end of the session can be reused for further analysis.

FLO-400 Information

FLO-400 is an essential laboratory device for 3D in situ hybridization and immunohistochemistry. It reduces the analysis time to 22 hours compared to 96 hours with a traditional tool.

PROBES

4 different probes can be used simultaneously (1 per Flow Cell). Baskets can be placed in order to multiply the number of embryos to be analysed.

RECYCLING

Recycling process: antibodies and RNA probes are recycled and can be much more diluted or reused for other experiments.

CONSTANT FLUX

Constant flow of reagent: 3D biological samples are constantly subjected to a solvent flow that ensures good conditions in the experiment.

The Flo400 is a safe, fast and easy-to-use research device that allows 3D samples to be analyzed using the in situ hybridization and immunohistochemistry (IHC) methods. Thanks to this equipment, it is possible to directly visualize the location and activation of genes of the cells concerned. The means used for this localization is in situ hybridization (HIS) of RNA probes (transcriptome analysis). It should be noted that the automaton also allows proteome analysis by immunohistochemistry, using antibodies instead of RNA probes.
